AB OBJECTIVE: To determine the functional activity of CD8 positive HIV-1
carrier cell clones. METHOD: A 4-hr two-color flow cytometric
cytotoxicity assay was used. The effector/target cell ratio analyzed was
in the 0.2-3.0/1 range. RESULTS, DISCUSSION AND CONCLUSION: Seven
HIV-1IIIB carrier CD8+ MOLT-4 clones from T-cell leukemia cell line
MOLT-4 with a CD4+CD8+ phenotype, and 5 CD8+ HIV-1 carrier cell lines
from 3 other CD4+CD8+ leukemia cell lines were used. Of the 7 MOLT-4/HIV
clones used, 4 exhibited cytotoxic activity specific to 2 immature T
cell lines, HPB-ALL and SUP-T1, of 21 clonal leukemia cell lines tested
with T-, B- or myelomonocytic origins. Pretreatment of the cytotoxic,
but not the noncytotoxic, clones with PMA augmented this specific
cytotoxic activity. Two-color cytotoxicity analysis indicated
unidirectional cytotoxicity; (i.e. CD8+-CD4+/CD8+). While no correlation
was found between cytotoxicity and 1) persistent infectious virus
release, 2) gp120 antigen or 3) p24 antigen expression, CD8 positivity
and its enhanced expression in the HIV-carrier clones appear to be
associated with specific cytotoxicity. The in vivo target cell
counterpart and the specific recognition molecules on the CD8-positive
HIV-1 carrier cells remain to be determined. The observation, however,
provides evidence for a mechanism underlying the immunodysfunction
associated with AIDS pathogenesis.
